研究概要

癌遺伝子は、細胞の増殖の制御にのみ働くのでなく、細胞の分化のプログラミングの段階で大きな役割をもつことが考えられる。本研究では、種々の特色ある分化系を用いて、機能、局在性の異なるいくつかの癌遺伝子の分化のプログラミングに果す役割を解明することを目標としている。
帯刀(研究代表者)は、マウス赤白血病細胞の分化誘導において、cーycが著名な発現変化をすることに注目し、人為操作可能な形でcーmyc遺伝子を導入し、強制的発現と分化の関係を解析した。その結果、cーmycは、この細胞の分化の決定の段階で、その発現量に依存して分化の頻度を決めることを明らかにした。また、cーmyc遺伝子の変異株の解析から、cーMycの癌化と分化の抑制に働く機能ドメインが異なることを示した。
大石班員は、同様の分化系において、分化誘導に関与する細胞内因子(DFーI,DFーII)がチロシンリン酸化により制御を受けていることを示し、チロシンリン酸化阻害剤が分化誘導に効果を表すことを示した。
石井班員は、cーMybが、転写因子として遺伝子上の特異的配列を認識して結合することを示すとともに、cーMybの機能ドメインが3つに分割できることを示した。
野田班員は、ras遺伝子が、褐色細胞腫PC12の神経細胞分化誘導を起すこと、そのときのシグナル伝達機構を解析した。
他の班員も、それぞれの特有の細胞分化系における癌遺伝子の働きを分子レベルで解析し、分化のプログラミングと直接癌遺伝子が結びつくことを示している。
