研究概要

サイクロトロンで産生される超短寿命のポジトロン核種^<11>C,^<13>N,^<18>Fおよび^<99m>Tc,^<123>Iを用いた新しいがん診断法の開発を目的として、各種標識化合物の合成とその体内挙動に関する検討、ポジトロンCT(PET)装置の開発とその動態機能解析法の開発に関する研究が行われた。
放射性薬剤については、癌化に伴う細胞膜の変化を利用する6-[^<18>F]フルオローLーフコーゼおよびN-[^<18>F]フルオロアセチルーDーグルコサミンを合成し、腫瘍親和性が認められた。また癌親和性グルコースー誘導体として、ヘキソキナーゼのグルコースリン酸化反応に関与するNー(mーヨードベンジル)ーDーグルコサミン(BGA)の膜透過性の向上を目的として、1,3,4,6ーテトラーOーアセチルーBGAが合成され、腫瘍における高摂取が認められた。また腫瘍親和性の[^<18>F]標識ヘキソースおよびalpha^<11>CH_<3->脂肪酸の合成法が確立された。またZn-62/Cu-62ゼネレータ・システムによる^<62>Cu標識放射性薬剤の開発の検索が行われた。
頭部用の高解像力ポジトロンCT装置が完成し、また、PETの臨床利用における解析法が検討され、臨床上の有用性が示された。
臨床研究では、脳腫瘍、肺癌、甲状腺癌患者に^<18>FDG,^<11>O-メチオニンなどを投与し、腫瘍への集積、治療による変動、他の医用画像との比較が行われ、腫瘍における特異な代謝の評価と、これを利用した腫瘍イメージングの有用性が示された。また腫瘍のRI摂取とMRスペクトロスコピィによるin vivo測定による組織のpH値との関係が検討され、腫瘍のRI摂取機序の解明に有用な方法になり得ることが示された。
本研究で示された成果は、さらに膜透過、酵素活性、受容体活性などの特異的な機能測定を目的とした新しい標識化合物の開発および臨床利用、高性能ポジトロンCT装置の開発による定量性の向上へと進展し、腫瘍の病態生理の解明、がん診断法の新しい展開が期待される。
