研究概要

本年度各班員の研究成果の大要を次項に記す。(i)シャトルベクターによるTrpーP2誘発突然変異の詳細な解析から、TrpーP2の変異一がん原性はDNAのグアニン附加体(C8GーTrpーP2)の生成による変異生成が主要であること、その誘発機構としてSlippage misalignmentモデルを提示した(加藤、木村)。この仮説を更に検証するため、AAFおよびOHーTrpーP2誘発変異の解析を行った。(ii)真核細胞でのこれら新しい知見を、データーの蓄積の多い大腸菌で比較するためIQおよびTrpーP2の変異スペクトルの解析を進めている(大西、尾川)。(iii)突然変異誘発とはDNA修復欠損の関係を調べるため、XP、AT細胞株にシャトルベクターを導入した系を樹立した。それを用いて、発がん物質により誘発された突然変異の解析を進めている。(古山)。(iv)シャトルベクター系では、cDNA遺伝子を用しているがゲノム遺伝子での研究も重要である。遺伝子増巾法(PCR法)の応用によってゲノム遺伝子の解析も可能となってきた。ヒトのゲノムHPRT遺伝子の突然変異をPCR法で解析する方法をほぼ確立した(加藤,松永)。立花らによって、これまでにサザンブロット法で解析されたヒト、リンパ芽球のHPRT変異細胞のゲノムHPRT遺伝子をPCR法で詳細に解析する実験がスタートした(巽)。(v)マウス培養細胞の修復欠損株を用いて、IQによる突然変異誘発頻度を調べたが、これまで高い感受性を示す修復欠損株は見つかっていない(佐藤)。
