癌転移に関与する細胞側要因の解析

研究概要	本班の目的は癌転移機構の解明に資する基礎的データの提供にある。竹市は細胞接着分子カドヘリンの発現が弱く、かつ不安定な高転移性マウス卵巣腫瘍株を用いてカドヘリン活性と転移能との関係を探る目的で、PカドヘリンcDNAの導入法としてβーアクチンプロモーターをつけてのelectroporation法が最適であることを見つけ、現在、同法によりPカドヘリン導入細胞の分離を試みている。谷口はsrcあるいはrasで形質転換したラット細胞にvーfos導入により観察される転移能増強は夫々運動能の増強を伴なう浸潤能の増強が、生体防御システムに対する感受性低下が一要因であることを明かにした。またB16黒色腫で見つけた新種アクチン(βm)が転移能抑制的に働くことをcDNA導入実験で示した。山本はヒト乳癌の病期、転移などの臨床データとcーerbBー2の過剰発現との関連を、乳癌症例50例を対象に解析した結果、その20%にcーerbBー2の増幅を伴うこと、38%にWestern blot法でErbBー2蛋白の過剰発現があること、免疫組織化学的に見てErbBー2蛋白は正常乳腺組織では発現は全くなく、癌組織においてのみ発現すること、またスキルス癌で最も高率に発現増大があり、病期、リンパ節転移の度合いとErbBー2蛋白増大との間に正の相関があることを明かにし、ErbBー2の発現増大は乳癌の進展を促進していることを示唆した。またこの発現増大は遺伝子増幅による場合と、転写/翻訳レベルでなされる場合のあることが明かにされた。横路は遺伝子でラベルしたマウス線維肉腫細胞を用いて継代早期より腫瘍内が1つの優性クローンにより占められることを報告してきたが、本年度はその肺転移巣も同じクローンにより形成されることを明かにした。その他、転移関連膜糖蛋白質に関する研究、Balb/c3T3の変異株を用いてのsrc導入による形質変化の研究、ヒト黒色腫微小転移形成の予防を目指したリコンビナント黒色腫ワクチンの研究などにも着実な進展が見られ、所期の目的を達成すべく順調に経過している。
