腫瘍局所に対する至適な化学療法の確立に関する研究

研究概要

癌性腹膜炎(胃癌、腸癌、卵巣癌、等)に対して、基礎及び臨床的治療研究で昨年に比べて大きく前進した。すなわち、我々の独自の研究によって開発された処のいわゆる昇圧2経路化学療法は、基礎実験的研究が大いに進展し、肝転移癌、子宮体部移植癌に対して局所動脈内DDP+ATーII投与、10分後にSTSを静注する時間差投与化学療法が極めて優秀な制癌性を発揮し、しかも腎毒性を強く抑制出来ることが明かとなった。
更に動物実験が進展して、遂に2経路投与に頼る事なく、同一の静脈より時間差をおいて投与(DDP+ATーII、5〜10分後にSTS)すれば、全身的薬剤投与によっても極めて優れた制癌効果を得ることが証明された。この治療法により臨床的に末期の卵巣癌、胃癌、腸癌等による癌性腹膜炎患者の多数例が治療された。高率に腹水減少、生存期間の延長等が観察され、現在、更に症例を増して検討続行中である。一方、臨床的肝癌、肝転移癌に対して栓塞化学療法が種々の工夫改良されながら症例を増加させている。
シスプラチンの各種誘導体についてその薬理動態、制癌性の優劣が比較検討され、制癌剤の高分子化について腫瘍内血管の透過性との関連から開発が研究された。
