研究概要

オカダ酸とデイノフィシストキシンー1等のオカダ酸クラスの発がんプロモーターに加えて、本年度は、カリキュリンAを新たに見い出した。更に、これらの作用機構を明らかにすることができた。オカダ酸クラスの発がんプロモーターは、細胞質内のプロテインフォスファターゼ1及び2Aに結合し、その脱リン酸化酵素活性を阻害した。脱リン酸化酵素活性の阻害と、プロテインフォスファターゼへの結合性は良く一致しているので、オカダ酸クラスプロモーターのレセプターは、プロテインフォスファターゼであることが証明された。オカダ酸分子のC1のカルボキシル基とC24の水酸基が水素結合をし、生じた空間が活性に重要であることが指摘された。アオコに含まれる肝臓毒、ミクロチスチンは、肝だけでなく、腎、大脳、小脳にも病理組織学的な障害を生じることが見い出された。発がんプロモーション活性と、アラキドン酸代謝亢進は密接な関係にあると考えられていた。しかし、活性化ras遺伝子を含む細胞では、アラキドン酸代謝は、むしろ、低下していた。この理由を更に検討している。
発がんプロモーターTPAについて、いくつか新しい結果も見い出された。TPAをマウス骨芽細胞に処理すると、4時間で、活性化される遺伝子が8個見い出され、その中に、メタロチオネインの遺伝子が含まれていた。TPAによりリン酸化される基質を検討した結果、分子量80kの2つの蛋白質が見い出された。その蛋白質の同定については、現在、検討中である。HLー60細胞内に、TPAタイププロモーターが特異的に結合する蛋白質を分離した。この蛋白質は、TPAタイププロモーターと結合後、核内に移動する等興味深い。
本年度の実績は、初年度であるにしては、活発な、又、重要な実験結果が得られた。
