gamma/delta型T細胞による抗腫瘍防御機構の解析と治療への応用

研究概要	T細胞はその抗原レセプターより、alpha beta型とgamma delta型の2種類に分類される。抗腫瘍免疫などの生体防御機構におけるgamma delta型T細胞の役割を検討するために、細胞内寄生性細菌であるListeria monocytogenesや、メチルコラントレン誘導のMeth A fibrosarcomaをマウスに移植した後に出現してくるgamma delta型T細胞について解析をおこなった。 (1)リステリア感染症……致死量以下リステリア菌を腹腔内に投与して1日目、3日目、8日目の腹腔内出T細胞を調べると3日目にCD48のgamma delta型T細胞が増殖しており、8日目になるとCD4またはCD8のalpha beta型T細胞が増加した。3日目に増加しているgamma/delta型T細胞の抗原特異性を検討したところ、Heatーkilled Listeriaは反応しなかったが、BCG由来のレコンビナント65kd Heat shock protein(HSP)で有意に増殖した。リステリア菌だけでなく感染3日目の腹腔マクロファージには65kd HSPとホモロジーの高いHSPが検出された。以上の結果より、リステリア感染早期に発現するgamma/delta型T細胞は、細菌または自己細胞の発現するHSPを認識して活性化されるものと考えられる。 (2)担癌状態……BALB/c由来のMeth A腫瘍を皮下に移植した同系BALB/cマウスの局所リンパ節で増加するT細胞を検討した。移植後3日目でCD4ーCD8 gamma delta T細胞が有意に増加しており、移植後8日、14日となるにつれて、CD4^+alpha beta T細胞が増加した。腹腔内でも同様に移植後早期にgamma/delta型T細胞が増殖し、14日目以降ではCD4^+細胞が増加した。VACー1に対する細胞障害活性は移植3日目で有意に高く、14日目ではむしろ低下していた。腹腔内で早期に増加するgamma/delta型T細胞は65kd HSPに反応した。一方、局所リンパ節ではgamma delta型T細胞のみならず、CD4^+T細胞もHSPに反応して増殖することがわかった。Winn assayでこれらのHSP特異的T細胞の抗腫瘍活性を調べたところ、有意に腫瘍の増殖をおさえた。
