制がん剤耐性獲得の多様な分子機構と耐性克服法の開発

研究概要	制がん剤耐性獲得に関与する機構の多様性を把握するために、我々は様々な制がん剤に対する耐性がん細胞株を樹立し、関与する耐性因子を検索した。同時に耐性因子を標的にした耐性克服法を開発する仕事を進めた。その結果、 1.多剤耐性遺伝子mdrー1の発現に重要なプロモーター及びエンハンサー領域を単離し、ビンクリスチンをはじめとする制がん剤によってmdrー1遺伝子の発現が促進することを見出した。 2.mdrー1遺伝子産物GP170を標的にしてジピリダモールならびに数多くのジハイドロピリジン系薬剤の中から培養系及び動物実験系で耐性克服する組合せを見出した。 3.シスプラチン耐性株を樹立した。耐性株において親株に較べグルタチオンSートランスフェラーゼ(GST)が数倍上昇していること、さらに感受性復帰変異株においてGST活性が再び元に戻っていることを観察した。 4.GSTーpiのcDNAを導入した株でGSTーpiが数倍上昇しており、2ー3倍のシスプラチン耐性を獲得することを見出した。 5.エトポシドVPー16やテニポシドVMー26に対する50ー100倍の耐性株を単離し、DNAトポイソメラーゼII及びVPー16/VMー26の透過性が変化していることを観察した。今後、mdrー1遺伝子のプロモーター領域に結合する転写因子を明らかにするとともに、制がん剤による転写調節機構を把握したい。また、シスプラチン耐性についてはGST以外の耐性因子の同定ならびにVPー16/VMー26耐性を左右するDNAトポイソメラーゼの役割についての解析を進めていきたい。
