前胸腺刺激ホルモンを中心とした昆虫脳ペプチド類の構造、機械および動態

研究概要

本年度の成果は以下のとうりである。
(1)カイコ前胸腺刺激ホルモン(PTTH)と相同性のある分子をPCR法を駆使することにより、エリ蚕からクロ-ニングした。カイコPTTHと同様、ペプチド内に7個のシステインが存在し、アミノ酸配列のホモロジ-は約50%であった。また、カイコPTTHでは糖鎖が付加しているアスパラギンはセリンに置換されていること、前駆体に含まれる2つのペプチドの配列はカイコPTTHとはかなり異なっていることが明らかとなった。また、エビガラスズメ脳中にもカイコPTTH抗体と反応するペプチドが存在することを免疫組織化学的手法を用いて明らかにした。
(2)カイコ羽化ホルモン(EH)の遺伝子構造を明らかにした。解析の結果、EHは、26残基のシグナルペプチドを含む88残基のペプチドとして翻訳されること、また、ゲノム上では3つのエクソンからなることが明らかとなった。さらに、in situハイブリダイゼ-ションおよび免疫組織化学により、EHは脳間部4個の神経分泌細胞で合成されていること、幼虫期には、軸索を経由して腹部にいたる中央神経系へ輸送され、蛹期には、主に側心体に貯蔵され、放出されていることが明らかとなった。
(3)カイコ性フェロモン生合成活性化ペプチドに対する抗体を作製し、免疫組織化学的手法により、食道下神経節中の12個の細胞で合成されていることを明らかにした。
(4)カイコPTTHを大腸菌により、ボンビキシンを化学合成により大量合成し、高次構造解析のためのNMRの測定を行った。その結果、PTTHについては詳細な解析を行うためには安定同位体で標識することが必要であること、ボンビキシンについては測定中に不溶化したため、測定溶媒の検討が必要であることがわかった。
