膜タンパク質の疎水性アンカ-としてのPIの生物学的意義

研究概要

12名の研究者により、本年度に以下のような研究成果が得られた。
1.生化学的側面の研究。PIPLCにより遊離されたカイコのアミノペプチダ-ゼN、ウシ肝5'-ヌクレオチダ-ゼを精製し、さらに5'-ヌクレオチダ-ゼのcDNAのクロ-ニングを行い、全アミノ酸配列の9割を確定し、C末疎水アミノ酸配列の部分をほぼ同定した。また昆虫培養細胞TN-368のβ-グルコシダ-ゼがPIアンカ-蛋白であることを発見した(池沢)。蛋白合成系における延長因子EF-1αがPIアンカ-により小胞体膜に結合していることを発見した(鬼頭)。ラット肝及びヒト胎盤5'-ヌクレオチダ-ゼのcDNAクロ-ニングを完了し、それぞれの全一次構造を決定し、これらの蛋白質C末端PI結合部位の同定並びにPI複合体の組成分析を完了した(池原)。補体制御蛋白MACIFのcDNAをクロ-ニングし、アミノ酸配列のC末部からPIアンカ-の蛋白であると結論した(富田)、ウサギ小腸のトレハラ-ゼに対するモノクロ-ナル抗体5標品を得た(武居)。PIアンカ-酵素であるアルカリ性ホスホジエステラ-ゼIが、酪酸ナトリウムによりKBIII細胞で高レベルに誘導されることを見出した(中林)。腫瘍マ-カ-のうちでCal25分子にPIアンカ-が存在する可能性を見出した(菰田)。
2.免疫学的または病理学的側面の研究。PI結合膜蛋白であるアロThyー1抗原の特徴的な免疫活性がPIPLCに対して高い感受性を示すThy-1抗原の特徴的な免疫活性がPIPLCに対して高い感受性を示すことから、この活性の発現にPI膜結合が重要な役割を担うことを明らかにした(中島)。赤血球の抗原HRF-20がPIアンカ-抗原であることを明らかにした(岡田)。発作性夜間血色素尿症PNHの異常がDAFあるいはHRF-20の欠損によるらしいことを発見した(藤田)。ヒトPNH患者でDAFが正常、IF5抗原欠損の特異な症例を発見した(田口)。アルカリ性ホスファタ-ゼの単クロ-ン抗体を樹立した(上出)。
