研究概要

黄化ヤエナリ芽生えを材料として、ベンジルアデニン結合セファロ-ス4Bアフィニティ-カラムを用いることにより、サイトカイニン結合蛋白質を単離精製した。この蛋白質は分子量が約20万で、分子量48,000と45,000の2種類のサブユニットから成っており、ベンジルアデニンに対するKd値は7.5×10^<-7>Mであった。また、ジベレリンA_3結合セファロ-ス4Bアフィニティ-カラムを用い、ジベレリン結合蛋白質を単離し、部分精製することができた。植物ホルモンでその発現が調節されている、グルタミン合成酵素、フェニ-ルアラニンアンモニアリア-ゼについては、それらの遺伝子を単離することができた。さらに、オ-キシンによって調節されるmRNAのcDNAクロ-ンを得ることができたので、この遺伝子についても単離できる見込みがたった。これらの遺伝子のプロモ-タ-領域と種々の植物ホルモン結合蛋白質との相互作用を調べる計画は、期間内に達成できる状況になってきた。
矮性遺伝的植物に関する研究では、トウモロコシD8が、ジベレリンに対して高い親和性を有するジベレリン結合蛋白質を欠損している可能性が明らかになり、さらに詳しく調べているところである。またイネ種子をメチル化剤で処理することにより、矮性体をいくつか得ることができたので、これらの矮性体と植物ホルモン結合蛋白質との関係を明らかにしていきたいと考えている。
植物ホルモンの作用については、細胞壁微小管配向がジベレリンとアブシジン酸で調節されることや、細胞の伸長生長が、細胞壁糖鎖認識抗体で阻害されることなどが明らかになってきた。
