研究概要

1)DNA多型の応用について.
血痕からの人獣鑑別、個人識別に関して超高変異性を検出しうるDNAプロ-ブを用いての検出法のほかVNTR領域をPCR法で増幅して判定する方法が完成した。この成果を親子鑑定、法科学的試料に実際に応用した結果非常に有効であり、実務鑑定への道が開けた。一方、法科学的試料にはさまざまなものがあるが、特に焼死体の血液などからDNAを抽出する場合、多くの問題がある。本研究では超遠心法によりDNAを抽出して、VNTR用プロ-ブにより多型を判定することが可能になった。パライン切片から抽出したDNAによる個人識別もPCR法を用いることにより性別、親子鑑定に関して応用可能になった。これまでシングルおよびマルチロ-カス上のDNA高変異領域の多型同定にフィンガ-プリント用プロ-ブやVNTR検出用プロ-ブが用いられてきたが、このほかにRFLPとして報告されてきたものが数多くある。そのうちヒトガンマグロブリン遺伝子領域に検出される多型形質についても検討を行った結果、親子鑑定に有効な多型性バンドが同定された。更にApoBー100gene,ALDH2alleles,GST1は疾患との相関を示す点で重要な遺伝マ-カ-である。これに関し検索を行った結果、遺伝疫学的観点からのDNAマ-カ-に新たな道が開けた。
2)蛋白・酵素の多型の応用について
FactorI,ClR,BFなどの蛋白は血清中に含まれ,日本人でも多型性変異を示す遺伝マ-カ-である。親子鑑定の際、父権肯定確率の計算の基礎となる遺伝子頻度は重要であるため、地方差、地理的勾配の点から検討を行った結果、FactorI,ClRについては東北地方から関西、九州にかけての勾配の存在が認められた。また、Orsomucoid,唾液アミラ-ゼ、赤血球抗原性についても、遺伝子頻度、検出法に新しい知見が得られた。
