研究概要

I.計画年度2年目の今年度、班として行った主な行事は:
1)脳細胞培養に関する勉強会(平成2年4月28日於早稲田大学人間総合科学研究センタ-)、
2)班会議(9月2日〜4日於山梨県清里・地方公務員共済組合「清翠荘」)、及び
3)生物物理学会第8会年会シンポジウム「生物時計に関与する原始視覚系の分子機構」(10月13日於九州大学)、である。
II.各自分担作業に精力的を注いだ。新たに得られたな主な結果は、下記の通りである。
1)ゾウリムシの静止膜電位は、昼深く(より陰性)で夜浅い(より陽性)。cGMP濃度は、昼濃く夜薄い。
2)ゾウリムシにも11シス型レチナ-ル-が存在することを確認し、また抗カエルロドプシンウサギ抗体と結合するロドプシン様蛋白質を純度約50%まで部分精製した。
3)ゾウリムシやアラスカエンドウの原形質膜にはATP・GTPともに結合するタンパク質(AGP)が存在し、特にアラスカエンドウでは赤色光照射により6種類のAGPのATP結合性に2〜3倍の上昇が見られた。
4)性的に成熟したゾウリムシにIP_3をinjectionすると、光パルス刺激と同様に接合活性が誘起された。
5)エンハンサ-トラップ法を用いて、キイロショウジョウバエの生物時計突然変異系統の分離を試みた。その結果、短周期型6系統(自由継続周期、22.9〜23.3h)及び長周期型4系統が得られた。
6)抗ウシロドプシン抗血清及び抗ニワトリアイオドプシンモノクロ-ン抗体を用いて、ニワトリ及びウズラ網膜の免疫組織化学を行い、a)cone外節がアイオドプシン抗体で染色される、b)視細胞outer limitting membraneのゴルジ体存在部位が染色されることなどを見た。
7)幼若ラット脳の視交叉上核から遊離の神経細胞を調製・長期培養を行った。培養3週間後細胞還流液中のバソプレシンを測定し、約25%の培養皿から日内変動を観察した。
