研究概要

1.ゾウリムシのcAMP・cGMP、及びprotein kinase Cの概日変化に関し、
(1)cAMP・cGMP濃度は、昼高く、夜低いこと、)この変化は、恒案でも観察されること、そして明案サイクル下では、光のオン・オフ時に一過的な変動が観察されること、
(2)PKCも基本的には、cAMPやcGMPと同じ変動を示すこと、
などが明らかになった。
2.ショウジョウバエの生物時計に関する分子遺伝学的研究研究遂行上のために、新たにショウジョウバエの概日リズム周期の異なる二つの突然変異体、Toki・Ritsuを分離し、その遺伝学的・時間生物学的性質を解析した。更に、エンハンサ-トラップ法によって無周期突然変異1系統を分離した。
3.鳥類・ほ乳類の概日時計の光情報伝達機構に関し、
(1)ウズラ眼内メラトニンは、明暗サイクル及び恒暗下で明確なリズムを示し、これが角膜上皮細胞分裂リズムに関与している可能性が高いこと,
(2)遺伝的に杆体細胞を欠損するC3Hと正常細胞のCBAマウスを用いて、白色光照射によりメラトニン合成抑制を調べた結果、両系統の感度閾値に約100倍の差が認められたこと、これは杆体細胞の有無によるものと考えられること、
などが明らかになった。
4.ほ乳動物の時計の座とされている視交叉上核に含まれるペプチド量を、酵素抗体法で定量し、
(1)視交叉上核腹外側で合成される血管収縮性ペプチド(VIP)とガストリン放出ペプチド(GRP)は、恒暗条件ではリズムを示さないが、光刺激に対して、VIPはレベルが下がり、GRPは上がること、
(2)バソプレシン(AVP)とソマトスタチン(SS)は恒暗でも明確な日周リズムを示し、SSのmRNAの量は恒暗下でct0で最大の濃度を、ct12で最小の濃度を示すこと、
などを明らかにした。
