粥状動脈硬化症の成因に関する基礎的研究

研究概要

平成3年度の研究は、ほぼ計画通り進んできたが、まず成果としては、遺伝的に動脈硬化を起こしてくるWHHLウサギを用いて、我々がin vitroで最もatherogenicリポ蛋白と考えていた酸化LDLの存在を証明することができた(Arteriosclerosis and Thrombosis 1991)。また、in vitroで酸化LDLの形成を抑制することが確認できた抗酸化剤であるprobucolをin vivoでその作用を確認する意味で、WHHLウサギに長期間投与し、検討するかたわら、動脈硬化の退縮効果も合わせて検討を加えた。長期間(16ケ月間)の投与により、動脈硬化の抑制効果が十分に認められることを明らかにした。しかも退縮効果が期待できるという成果も合わせて明らかにできた(Atherosclerosis 1992)。山本等は、WHHLウサギと、コントロ-ルウサギにおいて、WHHLウサギの動脈硬化巣から抽出したmRNAと、コントロ-ルウサギの同部位から抽出したmRNAを比較し、動脈硬化巣に発現している特異的なmRNAを同定したところ、トランスフェリンmRNAが特異的に増加していることを見い出した。また、長鎖脂肪酸の代謝に必須のLACSを単離精製し、その構造決定に成功した(JB 1992)。
平滑筋細胞増殖因子についても、その単離を進めているが、量的に極めて少なく、分子量約10KDのところに活性が認められることがわかってきた。また、平滑筋細胞遊走因子が今回新たに見い出され、その解析を進めることになった(Atherosclerosis 1991)。中性脂肪の分解に必須のリポタンパクリパ-ゼに関しては、その欠損患者の遺伝子解析を行い、その異常を明らかにした(JCI 1991)。
プロスタグランディンに関する研究成果として、プロスタグランディンE受容体の単離、精製に成功したことである(JBC 1992)。
