研究概要

ホルモンによる遺伝子発現制御機構について最も解析が進んでいるのは、ステロイドホルモンであるが、既に明らかにされているホルモン・レセプタ-と標的遺伝子との直接作用に加えて、間接的に作用し、転写促進することが判明した(市原・森)。cAMPによる遺伝子の転写促進に関して、グルカゴンとコルチコトロピン放出ホルモンは、それぞれセリン脱水酵素とプロオピオメラノコルチン遺伝子に対して、CRE(cAMP応答配列)を介して作用している可能性が高い(市原・井村)が、P450遺伝子のACTHによる転写促進は、CREを介さない別の機構によることが示唆された(藤井)。生体内で広範な作用を示すインスリンについては、市山はセリン:ピルビン酸アミノトランスフェラ-ゼを、田中は、解糖系の二つの鍵酵素を取り上げ、その作用機序の解明を行っている。田中は一時的な発現系では明らかにし得なかったインスリン応答配列を、トランスジェニックマウスの系で同定した。活性型ビタミンDのレセプタ-は、ステロイドホルモンレセプタ-ファミリ-であることが最近明らかにされている。黒木は、活性型ビタミンDが表皮細胞の分化を促進する機構を、尾形は、副甲状腺ホルモン遺伝子が活性型ビタミンDによって転写抑制されることを見出し、その機構を解明しようとしている。松本は、アンドロゲン依存性の乳癌細胞におけるFGF様物質の産生がこの細胞の増殖に関与していることを明らかにした。大場は、α_1酸性糖タンパ質遺伝子のT3応答配列について、橋本は、抗高脂血薬によるペルオキシソ-ム局在脂肪酸β酸化系酵素群の誘導機構を、杉本は、消化管ホルモンによる膵酵素分泌調節因子の発現調節機構を解析している。
以上のように、従来の知見を深めるにとどまらず、新しい作用機序が明らかになりつつある。これらの研究をさらに進展させるために、重点領域研究として申請を計画している。
