研究概要	卵黄とグリコ-ゲン顆粒の多いカイコ卵において、メッセンジャ-RNA(mRNA)抽出にはセシウムTFA法が効果的であることを見出した。これによりノ-ザンブロットにおけるハイブリダイゼション能およびウサギ網状赤血球由来の無細胞タンパク質合成系における翻訳能が著しく上昇したので、以後は同方法を適用した。種々の時期にあるカイコ卵からmRNAを抽出し、無細胞系における翻訳産物を2次元ゲル電気泳動後フルオログラフィ-を作製することにより、翻訳可能なmRNAの組成の変動を詳細に追及した。カイコは卵期に胚休眠を行うことによって越冬する。休眠は胚発生初期の器官形成開始直前の状態で起こり、また、卵には母体由来のmRNAが大量に取り込まれているので、その保持(利用)と休眠との関係が問題となる。本年度の研究により、休眠前後では基本的なmRNA組成に大きな変化がないことが明らかとなった。従って、休眠中には母性mRNAがインタクトに保持されている可能性がある。そこで、胚発生・休眠中にmRNAがどの程度利用され、どの様な速度で代謝されるかを追及する目的で、さしあたり、mRNA全般の動態を調べるのみでなく、少数の特異的mRNA分子種に着目して研究を進める方策をも検討した。このための最適な分子指標として、解糖系の酵素であるアルドラ-ゼに着目した。酵素は活性によってその存在が検索できる点で便利である上、アルドラ-ゼは一般にアイソザイムの存在が知られているので、多型が存在する可能性がある。九州大学に保存されている種々のカイコの突然変異系統を用いてアルドラ-ゼアイソザイムを解析したところ、胚のアルドラ-ゼに関して系統間多型があることを見出した。従って交雑次代におけるmRNAの指標として利用できる可能性があり、母性mRNAの区別に利用できる。アルドラ-ゼは母体体液にも存在していることをも見出したので、世代循環を追及する系としても役立つことが示唆される。
