研究概要

本研究は、ウイルスの増殖に必要な遺伝子に対するアンチセンスRNAを発現するトランスジェニック動物を作製し、当該ウイルス抵抗性品種の作出に関して検討することを目的としている。今年度は以下のような成績を得た。
1.作製されたマウス肝炎ウイルス(MHV)のヌクレオキャフシド(N)タンパク質の遺伝子に対するアンチセンスRNAを発現するトランスジェニックマウスの家系について感染実験を行った。すなわち、出生後1〜2日の新生児に600〜1.000PFUのMHVを感染させ、その生存率を検討したところ、対照の非トランスジェニックマウスでは感染後約3日目から死に始め5日目における生存率は20%程度であった。これに対し、2系統のトランスジェニックマウスの内1系統では明らかに生存率の改善がみられ、7日後でも約50%の生存率が得られ、8日以後の死亡は認められなかった。この成績はMHVのNタンパク遺伝子に対するアンチセンスRNAを発現するトランスジェニックマウスがMHVの致死的感染に対して抵抗性を有することを示した。今後、致死的感染に対するトランスジェニックマウスにおける抵抗性の発現機構やウイルス増殖の仰制における分子生物学的あるいは病理学的解析などが検討課題として考えられるが、本研究で得られた成績からアンチセンスRNAの発現によるいわゆる細胞内免疫によって個体に当該ウイルスに対する抵抗性を賦与できることが示され、家蓄・家禽へのその応用の可能性を強く示唆する成果を得た。
2.より効率よくウイルス遺伝子の発現を仰制するためにヒトのEF1プロモーターなど各種のプロモーターあるいはリボザイムの応用についても検討を行い培養細胞を使用して効果的にMHVの増殖を示すことが示され、今後、導入遺伝子を改善するための知見が得られた。
