研究概要	カルシウムイオン/カルモデュリン依存性の多機能性蛋白質燐酸化酵素であるカルモデュリン依存蛋白質燐酸化酵素II(キナ-ゼII)は、カルシウムイオンによる生体機能の調節、特に中枢神経の機能発現とその調節に中心的な役割を演じていると考えられている。キナ-ゼIIの生理的な役割を明らかにするために、本年度は主としてキナ-ゼIIの構造と機能の関連に関する研究、キナ-ゼIIによるチロシン水酸化酵素の活性化機構、新たな脳に特異的に分布するカルモデュリン依存性蛋白質燐酸化酵素IV(キナ-ゼIV)の研究を進めた。キナ-ゼIIはN末端から286番目のアミノ酸であるスレオニンを自己燐酸化して活性を発現するが、この時キナ-ゼIIはカルシウムイオン/カルモデュリン非依存性活性をも発現する。この286番目のスレオニンを燐酸化した酵素の性質を反応速度論的に解析した。ド-パミン、ノルアドレナリンなどの神経伝達物質の合成速度を決めているチロシン水酸化酵素は、キナ-ゼII、サイクリックAMP依存性蛋白質燐酸化酵素(Aキナ-ゼ)、プロテインキナ-ゼC(Cキナ-ゼ)の3つの代表的なセカンドメッセンジャ-応答性の多機能性蛋白質燐酸化酵素によって燐酸化される。それぞれのキナ-ゼによるチロシン水酸化酵素の燐酸化サイトと燐酸化量を調べ、チロシン水酸化酵素の活性化機構を考察した。脳に特異的に分布するキナ-ゼIVをラットの小脳から精製し、その性質を調べた結果、基質特異性の広い多機能性蛋白質燐酸化酵素であることが明らかになった。これはAキナ-ゼ、Cキナ-ゼ、キナ-ゼIIに次ぐ4番目のセカンドメッセンジャ-応答性の多機能性蛋白質燐酸化酵素で、脳で重要な働きをしていることが考えられる。キナ-ゼIVは自己燐酸化反応によって自己を活性化するが、Aキナ-ゼによって燐酸化されると逆に不活性化されるという興味深い性質が明らかになった。
