ねじれたπ電子系円二色性スペクトル法の開発と天然有機化合物の絶対構造決定への応用

研究概要

有機化合物の絶対構造を非経験的に決定する方法として、X線法とならんで円二色性(DC)スペクトル法が有用である。我々は「CD励起子カイラリティ-法」の拡張として「ねじれたπ電子共役系」の絶対構造決定にπ電子SCFーCIーDV分子軌道法によるCDスペクトルの理論計算が有用であることを見いだし、天然物に適用して絶対構造を決定してきた。本年度はCD法とX線法を組み合わせて以下の研究成果を得た。
(1)天然物のアトロ-プ異性:CDスペクトルの理論計算により長年の間未決定であった天然の回転異性体であるビフラボンの絶対立体化学を初めて決定した。
(2)ビスシアニン色素の可視とCDスペクトルの異常に大きな励起子相互作用:励起子相互作用はCDでは容易に観測されるものの、UVではほとんど観測されていなかったが、我々はビスシアニン色素のUVで70nmもの異常に大きな励起子分裂を初めて見いだし、励起子理論を確立した。また理論計算により、その機構を明らかにした。
(3)CD励起子法による鎖状ポリオ-ル系の絶対構造決定:光学活性鎖状1,2ー、1,3ーグリコ-ルおよび1,2,3ートリオ-ル類へCD励起子法を適用し、絶対配置を決定できることを明らかにした。また配座解析から機構を明らかにした。
(4)光屈性制御物質ラファヌサニン類の構造の訂正:植物の光屈性の機構については論争が展開されているが、我々は光屈性制御物質ラファヌサニン類の構造が間違っていたことを見いだし訂正した。なおラファヌサニンは当初、光学活性と報告されていたが、ラセミ体である。
(5)12位に水酸基を持つネオクレロダン系ジテルペンの絶対立体化学。
(6)放線菌の生産するアセチルコリンエステラ-ゼ(AChE)活性阻害物質NK901093の構造と絶対配置を決定した。
