ショウジョウバエ集団における適応進化の分子集団遺伝学的研究

研究概要	1.ショウジョウバエGpdhに遺伝子座のDNA変異の調査(高野) Gpdh遺伝子座の二つのアロザイム多型、FとSについて構造遺伝子を含む5690bpの領域の塩基配列を決定した。またオナジショウジョウバエについてもこの領域の塩基配列を決定し、FとSの間の分岐時間を推定した。この結果Gpdh遺伝子のFーS間の分岐時間はAdh遺伝子座のFーSの分岐とほぼ同じ時期におこったこと、キイロショウジョウバエ種内のDNA多型の分布はエクソンの同義座位においてもっとも多型の度合が大きく,イントロンや非翻訳領域の2倍に達しており、これはAdhで得られた結果と一致することが明らかになった。 2.トランスポゾンの転移率の推定(原田) トランスポゾンの転移は複雑な突然変異を引きおこすが、これらが適応進化にどのように貢献しているのかを検討するにあたり、重要なパラメタ-である転移率の推定を試みた。自然突然変異を約400世代蓄積した系統AW,JHについてcopia様因子(copia,412,17,6)およびLINE様因子(I,Jockey),hobo因子のDNAをプロ-ブとしてin situハイブリダイゼ-ション法によりそれぞれの挿入部位を決定した。転移はCopia様因子で4.8×10^<-5>/世代と推定され、欠夫はcopia様因子およびLINE様因子ともに非常に低く、2.5×10^<-6>/世代であった。I,hobo因子で比較的高い転移があった。 3.P因子の集団遺伝学(山崎) 完全型P因子(2.9kb)と欠夫型P因子(0.4ー2.8kb)の集団内分布をサザンハイブリダイゼ-ション法で調べた。調査したのは石垣、赤湯、大阪の集団である。この結果完全型P因子の分布に集団間の違いがあること、どの集団でもKP因子と思われる0.9ー1.1kbのP因子が全体のコピ-数の半数以上を占めていることがわかった。
