侵害受容被殻説の検証

研究概要	視床腹側基底核群被殻領域に分布する特異的侵害受容ニュ-ロンと広作動域ニュ-ロンにおける痛覚情報のシナプス伝達機構を明らかにする目的で、上行性疼痛抑制系の在存を確かめる研究を行なった。ウレタン・クロラロ-ズで麻酔したネコの中脳の背側縫線核(NRD)とそれに隣接する中心灰白質(PAG)腹内側部に条件刺激を加えると、腹側基底核群に属する後外側腹側核の被殻領域に分布する特異的侵害受容ニュ-ロンと広作動域ニュ-ロンの内臓求心性線維刺激(試験刺激)に対する反応が抑制された。中脳を出て、延髄腹内側部(主として大縫線核)で中継された後、後側索を通って脊髄後角に下行する下行性疼痛抑制系を頸髄の高さで遮断しても、条件刺激による抑制は消失しなかった。また頚髄の高さで脊髄前側索を刺激したときに誘発される短潜時スパイク発射も条件刺激によって抑制された。これらの結果から中脳のNROと腹内側PAGから視床腹側基底核群に向かう上行性疼痛抑制系の存在が見出された。延髄大縫線核に条件刺激を加えても同様な抑制がみられた。この場合の抑制は、大縫線核に線維を送るNRDおよびPAGニュ-ロンの逆方向性興奮によることが見出された。視床髄板内核群に分布する侵害受容ニュ-ロンについても同様な実験を行なったところ、一部のニュ-ロンで抑制がみられ、上行性抑制機序の関与が証明された。しかし、一部のニュ-ロンはNRD、PAG刺激に反応して興奮した。また、大多数のニュ-ロンは影響を受けなかった。この結果は、NRD、PAGを出る疼痛抑制系は、主として腹側基底核を中継核とする痛覚系に作用し、髄板内核群を含む情動系に対する抑制作用は弱いことが明らかとなった。モルヒネの作用点についても痛覚系と情動系で異なることが浮き彫りにされた。
