クライオカルチャ-、セルブロット法による神経成長・標的認識機構の分子生物学的研究

研究概要	神経細胞の軸索成長と標的認識の分子機構の解明は、発生期における回路形成、シナプス発芽や再生の機構の解明に不可欠な極めて重要な問題である。この問題の解明に有力な手段の1つは、脳内に起こるこれらの現象を再現するin vitro標本を用いることである。しかし通常用いられる培養系では脳の構造との関係を知ることが困難である。本研究では、クライオカルチャ-法及びセルブロット法という2つの新しいin vitroアッセイ系を確立して、発生、発芽や再生などの現象に関与する分子を捉えることを目的としておこなった。本年度は初年度におこなったクライオカルチャ-法によって得られた細胞接着・突起伸展の抑制現象に関与する知見をまとめて論文として発表した(Neuroscience Research 8,1990)。さらにそれに引き続いてこの因子の精製を進めた。そのためにラットの脊髄切片から各種の可溶化剤を用いて抑制活性を抽出し、これを抗原としてモノクロ-ナル抗体の作製を試みた。その結果、切片の白質への細胞の接着抑制を低減させる抗体が得られた。いっぽう、この因子の性質を明らかにするために、各種のレクチンをラット脊髄切片に投与することによって白質への細胞接着の変化を調べた。用いた15種類のレクチンのうちConcanavalin Aのみが白質への細胞接着抑制活性を著しく低減させた。また同様の現象はサクシニルConAによっても観察され、レクチンの効果はαーメチルマンノシドの投与によって消失した。以上のことは、この因子はConAによって認識される糖鎖をもっていることを示している。
