ミエリン形成障害ミュ-タントにおけるオリゴデンドロサイト変性脱落の機構の解明

研究概要	遺伝性にミエリン形成障害を示すジンピ-マウスにおいてはミエリンの主要構成蛋白質であるプロテオリピド蛋白(PLP)の遺伝子内に点突然変異が見い出されており、ミエリン形成細胞であるオリゴデンドロサイト(ODC)が変性脱落する。本研究において、遺伝子導入を通してその治療を試みた。まず、ジンピ-変異を同定する遺伝子診断法を開発した。すなわち、ジンピ-変異の両側をはさむようにオリゴヌクレオチドを合成し、その領域をPolymerase Chain Reaction(PCR)法により増幅した。ポリアクリルアミド電気泳動後メンブランにトランスファ-し、オリゴヌクレオチドプロ-ブで検出することにより、野生型とジンピ-型の識別ができるようになった。次に、PLPの発現ベクタ-を用いてトランスジェニックマウスを作製した。マウスPLP遺伝子のプロモ-タ-領域とポリA付加部位を持ち、間にラットPLP cDNAを含んだPLPミニジ-ンを構築し、マウス受精卵前核に注入した。生まれて来た24匹のトランスジェニックマウスを解析したが外来性のPLPミニジ-ン由来のPLPを発現しているマウスはいなかった。この理由としてPLP発現ベクタ-に発現に必須な領域を充分に含んでいなかったことが考えられるのでPLP全遺伝子をクロ-ニングし、トランスジェニックマウスを作製し直すことを計画した。Lorist Bコスミドベクタ-を用いてマウスジェノミックライブラリ-を作製しPLP cDNAおよびPLP遺伝子の5′、3′末端に対応する合成オリゴヌクレオチドでスクリ-ニングしたところ、5′上流領域20kb、3′下流領域5kbを含んだPLP全遺伝子をクロ-ニングすることに成功した。現在、このものを用いてトランスジェニックマウスを作製中である。この系が確立されれば導入するPLP遺伝子を改変することによりPLP異常とオリゴデンドロサイトの変性脱落との関連が推察できると思われる。
