細胞増殖因子によるホスファチジルイノシト-ルキナ-ゼ活性化機構の解明

研究概要

PDGFやCSFー1の様な細胞増殖因子が受容体の待つチロシンキナ-ゼを活性させ、標的蛋白のチロシン残基をリン酸化させることで情報を核に伝え、細胞増殖を促していることが分かっている。その標的蛋白の1つとしてホスファチジルイノシト-ルキナ-ゼが見つかり、チロシンキナ-ゼ系とイノシト-ルリン脂質情報伝達系の接点として注目を集めている。ホスファチジルイノシト-ルキナ-ゼの細胞増殖への役割を明らかにする為、ホスファチジルイノシト-ルキナ-ゼをラット脳より精製し、cDNAをクロ-ニングした。ラット脳膜分画ホスファチジルイノシト-ルキナ-ゼは分子量85Kでイノシト-ルの4位の位置にリン酸を付加するP14キナ-ゼであった。ラット脳のcDNAライブラリ-により上記P1キナ-ゼをコ-ドするcDNAをクロ-ニングした。大腸菌発現システムに組み込み蛋白を合成し、モルモットで抗体を作成した。この抗体はラット脳上記P1キナ-ゼを認識した。
