肝炎高発系LECラットにおける劇症肝炎発症機構の免疫遺伝学および免疫病理学的研究

研究概要	LECラットの肝炎は常染色体劣性遺伝形式で発症し、また肝組織中への銅の蓄積、血清銅および血清セルロプラスミン値の低下、病理組織学的には巨大核を持つ巨大肝細胞の出現、肝細胞壊死などの特徴があり、ヒトのWilson病ときわめて類似した所見を示している。しかしながら、胆汁うっ滞などにおいても肝組織中への銅が蓄積することが知られている。そこで、我々はLECラットの肝臓への銅の蓄積が肝炎の結果であるのか、あるいは原因となっているのかを、ヒトWilson病の治療薬であるdーpenicillamineをLECラットに投与し検討した。その結果、dーpenicillamineの投与によりLECの肝炎は完全に抑制され、肝組織中の銅含量も著明に低下していた。dーpenicillamineの作用機作として、銅をキレ-トとし尿中に排泄することにより、肝組織中への銅の蓄積を防ぐことが知られている。すなわち、dーpenicillamineをLECラットに投与することにより、銅がキレ-トされ排泄された結果、肝組織中への銅の蓄積が低下し、肝炎の発症が抑制されたと考えられる。このことより銅の肝組織への蓄積がLECラットの肝炎の原因であることが示された。今後、LECラットはヒトWilson病の発症メカニズム、特に銅代謝異常の解析に重要なモデル動物になりうるものと考える。
