好中球の活性酸素放出機構の解析とその細菌感染防御における役割

研究概要	細菌感染に際し宿主が動員する好中球、マクロファ-ジ等の食細胞は酸素を還元して毒性の強い活性酸素に変換する。この機能を遺伝的に欠く疾患では効果的な殺菌が行えず、重篤な感染を繰り返すことがわかっている。感染抵抗性の維持には、これら食細胞の正常な活性酸素放出能の維持発現が必須であると考えられる。我々は既にこれらの細胞が刺激により放出するのはスパ-オキシドアニオン(O2-)で他の酸素分子種は二次的に生じることを独自の方法を開発して明らかにしてきた。昨年度は、食細胞の形質膜に存在しこれに直接関与すると考えられる特別なシトクロ-ムの性質を明らかにするため、このチトクロ-ムの各部に対する抗体の分離に成功した。本年度は引続き、まず好中球の活性酸素産生の放出機構について解析を進めた。まずこの活性に関与すると考えられる形質膜上の特別なチトクロ-ムについて種々の部分を識別する抗体を用い、その存在様式について調べこれらが膜貫通性の蛋白質で形質膜に存在することを明らかにした。またマクロファ-ジを中心にその分布の臓器特異性について調査したところ、マクロファ-ジ系の細胞と考えられている脳のミクログリアおよび臍帯のホフバウエル細胞の他、Bリンパ球にも存在することを見い出した。なおホフバウエル細胞を用い、CGDの産前診断法を確立した。一方末梢血中の好中球が放出する活性酸素のレベルを血液化学発光として測定し、その測定が重傷熱傷患者の好中球機能とオプソニン活性をモニタ-するのに極めて有用であることを示した。これにより同種の患者の救命率が格段に向上した。
