ワクチンの免疫力増強に有用な免疫アジュバントの開発

研究概要	ヒトの感染症に対する種々のワクチンの開発研究が望まれるところであるが、それらワクチンの効果発現には強い免疫アジュバントの併用が必須であろうと考えられる。本研究ではその応用を考慮し、種々の合成免疫アジュバント、特にアシル・ムラミルジペプチド(MDP)誘導体、トレハロ-スマイコレ-ト(TDM)誘導体あるいはキチン誘導体などを化学合成し、それらの免疫増強活性を指標にスクリ-ニングし、副作用が少なく応用可能な免疫アジュバントを検索した。TDMは元来結核菌から精製された毒性糖脂質であることから、種々のTDM関連化合物の毒性試験も行なった。TDMは著明な体重減少を伴ない2週間以内に全例死亡した。その他の関連化合物は致死活性を示さなかった。単糖型のTDN関連化合物のなかで、GIcNAcM(1-deoxy)[6位にミコ-ル酸(M)が結合し更に1位をデオキシ化したGIcNAc]は強くマクロファ-ジを活性化し殺腫瘍性を誘導した。更に致死毒性及び体重減少も認められなかった。以上の成績から、糖部分の化学修飾によりその生物(免疫)活性が著明に変化することが明らかとなった。更にGIcNAcM(1-deoxy)は転移性腫瘍の移植局所近傍に投与した場合に、原発腫瘍の大きさ(増殖)には変化が認められなかったが、その肺への転移を有意に抑制した。一方、TDMを投与した場合も著明な転移抑制効果を示したが、著しい体重の減少を伴ない死亡例も観察された。以上のスクリ-ニングの成績より極めて低毒性の合成免疫アジュバントの候補が見出された。今後更にその他の評価系にて性状を明らかにすると共に、その投与形態などを考慮することにより感染防御効果を検討したい。
