リンパ球増殖分化過程におけるILー2受容体α鎖、β鎖発現の意義

研究概要

ヒト胎児由来の胸腺細胞におけるILー2受容体(ILー2R)α鎖、β鎖の発現状況を単クロン抗体を用いた2色フロ-サイトメトリ-法で調べた。その結果、CD3-CD4-CD8-pro T細胞ではILー2Rα鎖、β鎖は共に陰性であった。pro T細胞の中で、TCRα+β+CD4-CD8-細胞(DN細胞)の殆どがILー2Rβ鎖陽性で、一部の細胞では強いILー2Rα鎖発現がみられた。さらに分化したCD3du11+CD4+CD8+細胞(BP細胞)でもILー2Rβ鎖が陽性で、一部の細胞では強いILー2Rα鎖がみられた。しかし、CD3bright+DP細胞ではILー2Rα鎖、β鎖が共に陰性であった。また、胸腺細胞の中にもNK様細胞と考えられるNKHー1+細胞が存在し、この細胞では強いIL-2Rβ鎖の発現に加え、α鎖の発現もみられた。この細胞は末梢NK細胞とはILー2Rα鎖の発現において異っていた。胸腺細胞の中のCD4+T細胞は末梢CD4+T細胞の場合と同様にILー2Rα鎖は一部の細胞で陽性であるがβ鎖は陰性であった。逆にCD8+T細胞も末梢由来のCD8+T細胞と同様にβ鎖は陽性であるが、α鎖は陰性であった。以上の結果からclonal deletionに関与していると考えられている胸腺DP細胞にILー2Rα鎖、β鎖が共に発現していることが明らかとなった。すなわち、これら細胞のapoputosisにILー2/ILー2Rが何らかの役割を担っている可能性が考えられる。他方、T細胞分化と密接に関係する自己免疫疾患(SLE,PSS,RA)の患者末梢血細胞のILー2Rα鎖、β鎖発現を健常人の場合と比較したところ、これら活動期の患者のCD4+T細胞にα鎖とβ鎖の発現が共にみられた。さらに、SLEではB細胞の一部にILー2Rβ鎖陽性細胞が認められた。今後、これら自己免疫疾患におけるILー2Rα鎖、β鎖発現細胞の機能を解析し、疾患における意義について検討する。
