心臓機能の受容体調節異常におけるアラキドン酸代謝の役割

研究概要

心筋細胞;報告者は単一心筋細胞に於てアラキドン酸のリポキシゲナ-ゼ代謝産物がGタンパク制御Kチャンネル系を活性化することを発見した。その後、この新しい心筋Kチャンネル調節系がαアドレナリン受容体によるGタンパク制御Kチャンネルの活性化に際し働くことを報告した。また、フォスフォリパ-ゼA2を活性化するメリチンも同様にこのKチャンネルを活性化した。即ちアラキドン酸代謝物によるKチャンネルの活性化機構は種々の受容体依存性あるいは非依存性の新しい細胞内伝達機構として作用しうる。しかしながらその作用機構には未だ不明な点が多く、現在検討中である。また、従来のパッチ電極を用いた方法では心筋細胞内の生理的環境を長く安定した状態に維持することは困難であった。これはイオンチャンネル調節における細胞内代謝産物の関与の研究に際しての大きな限界であった。報告者はイオノフォア(ナイスタチン)を使用することにより細胞内環境を安定に維持できる方法を開発した。現在この方法と従来のパッチクランプ法を併用し、アラキドン酸化代謝産物の心筋細胞イオンチャンネルに対する効果と機序について検討を加えている。
平滑筋細胞;報告者はラット大動脈平滑筋細胞を用い、エイコサノイド系ならびにフォスフォリパ-ゼA2・Cの細胞増殖に与える影響を検討し、報告した。内因性降圧系プロスタグランジンは細胞増殖のG1休止期に産生が最大となり、G1休止期を維持し、細胞増殖を抑制した。高血圧自然発症ラットでは内因性降圧系プロスタグランジン産生が低下し、G1休止期が短縮し、細胞増殖速度が亢進することがみられた。今後、内因性プロスタグランジン産生異常の機序に関して、フォスフォリパ-ゼ誘導の遺伝子学的調節障害の側面から検討を加える予定である。
