T細胞の表皮親和性発現の機序の解明

研究概要	T細胞の表皮親和性発現の機序を明らかにするためにin vitro,in vivoにおいて表皮向性遊走を示す数種のT細胞クロ-ンを用いて主にin vitroで解析を行い以下の結果を得た。 1)表皮ケラチノサイトが産生することが明らかになっているサイトカインのILー1、3、6、GMーCSFを用いて表皮向性T細胞クロ-ンが、どのサイトカイに対して増殖し得るかを検討した。restingの状態ではどのサイトカインに対しても反応性を示さなかったが、抗原やMitogen刺激の表皮向性T細胞クロ-ンは、どれもGMーCSF、ILー6に対して反応性を示した。非表皮向性クロ-ンのうちのいくつかも同様にGMーCSF、ILー6に対する反応性を示したが多くのものは、反応性を示さないかあるいは、きわめて弱い反応性を示したのみであった。このことは表皮由来サイトカインのうちGMーCSFとILー6がT細胞の表皮向性を決定する大きな要素となっていることがわかった。 2)表皮向性、非表皮向性の種々のT細胞クロ-ンの細胞表面に発現している分子のうちcell adhesionに関係するCD2、CD4、CD3、LFAー1、CD8、Macー1についてflow cytonetryにて検討を行った。LFAー1分子が比較的選択的に表皮向性クロ-ン高濃度で発現しており、ある程度LFAー1分子の濃度とT細胞の表皮向性遊走能は平行していることがわかった。 3)表皮向性クロ-ンは、in vitroのagar plateを用いたmigretion assayにおいても表皮fragmentに対し遊走活性を示すが、抗LEAー1抗体の添加によりこの活性は完全に阻害された。divalentだけでなくFab fractionにも同様の遊走活性阻害能があった。二価のイオンに依存性であり、ヘプチド依存性の結果も、このin vitroの表皮向性遊走に、細胞骨格とassociateしたLFAー1分子が大きな役割を果していることを示している。
