I型糖尿病発症機序の解明:膵島細胞におけるMHC抗原誘導の分子機構とその制御

研究概要	〔目的〕I型(インスリン依存型)糖尿病の発症において、膵島細胞における主要組織適合性複合体(MHC)抗原発現の意義を明らかにすることを目的とした。〔方法〕ラットインスリノ-マ細胞株(RINm5F)を、種々のサイトカイン存在下で培養した後、RNAを抽出し、Northern blottingによってMHCクラスI、クラスIIおよびインスリン遺伝子の発現様式を検討した。用いたサイトカインは、ラットインタ-フェロンγ(IFNγ)、ヒト腫瘍壊死因子α(TNFα)、ヒトインタ-ロイキン1α(IL-1α)、マウスインタ-ロイキン4(IL-4)である。〔成績〕RINm5F細胞をIFNγ、IFNγ+TNFαの存在下で培養すると、MHCクラスIのmRNAは著明に増加したが、クラスIIのmRNAは誘導されなかった。IL-1αとIL-4は、クラスIクラスIIいずれのmRNAにも影響を及ぼさなかった。一方、インスリンのmRNAは、IFNγ、IFNγ+TNFαにより減少し、IL-1αでは増加傾向を示した。IL-4は、インスリンmRNAに影響を及ぼさなかった。〔結論〕ラットインスリノ-マ細胞株を用いた検討により、種々のサイトカインが、膵島細胞においてMHC、インスリン遺伝子の発現に影響を及ぼすことが示された。この事実は、これらのサイトカインが膵島細胞のMHC発現に関与し、増強ないし異常発現したMHC抗原が、I型糖尿病の発症に病因的意義を有することを示唆する。
