チロシン水酸化酵素遺伝子の培養細胞への導入:パ-キンソンモデルラットへの脳内移植

研究概要

申請者らは脳内移植によるパ-キンソン病症状改善を機序を解明することを目的として、カテコ-ルアミン生合成の律速酵素であるチロシン水酸化酵素(TH)cDNAを各種培養細胞へ導入し、パ-キンソンモデルラット脳内へ移植する試みを行った。昨年度は腫瘍細胞であるC6グリオ-マ細胞へ遺伝子導入を行ない、本細胞でLーDOPAが合成分泌されることを明らかにした。しかし、この細胞は腫瘍細胞であることから脳内移植実験には適さず、今年度は正常の線維芽細胞(NRK細胞)への遺伝子導入を試みた。免疫組織化学的検索および酵素活性の測定の結果NRK細胞においても活性のあるTH蛋白が発現されていることが示された。しかし、LーDOPAの合成分泌が見られなかったことから培地中にテトラヒドロビオプテリン(BH_4)を添加すると、LーDOPAの合成分泌が確認された。NRK細胞をラット脳内へ移植した場合にもBH_4脳内投与してやるとLーDOPAの遊離が観察された。この実験系を用いモデルラットの異常回転運動に対する効果を検討したところ、BH_4の添加により一過性に異常回転運動が改善されることが明かとなった。
これらの結果より遺伝子導入培養細胞は脳内移植の適切なドナ-細胞となり得ること、また、パ-キンソン病モデルラットの機能回復には必ずしも神経回路網の再構築は必須ではないことが示唆された。
