培養歯髄細胞におけるオピオイドペプチドの産出機序に関する研究

研究概要	平成3年度の研究目的はラット切歯歯髄の培養線医芽細胞を用いてブラジキニン(BK)ならびにその酸素分解産物des‐Arg^9‐BK(B_1)およびアルギニン(Arg)のエンケファリン(EK)産生酵素カテプシンBの活性化の機序を細胞内情報伝達機構との関連において比較検討することであった。(1)先ず、百日咳毒素(IAP)の影響を検討した。BKおよびB_1のカテプシンB活性化作用はいずれも細胞をIAPで前処理することにより抑制されたが、Argの作用はIAP前処理によっては影響されなかった。(2)キョ-トルフィン(Tyr‐Arg)のアンタゴニストであるLeu‐Argを用いてカテプシンBの活性化に対する影響を検討した。BKおよびB_1の作用はLeu‐Arg併用によって影響されなかったが、Argの作用は抑制された。(3)NO生合成を阻害する薬物L‐NMMAを用いてカテプシンB活性化に対する影響を検討した。BKおよびB_1のカテプシンB活性化作用はL‐NMMA併用によって影響されなかったが、Argの作用は抑制された。(4)フォスフォリパ-ゼCの阻害剤neomycin Bの影響を検討した。BK、B_1、およびArgのカテプシンB活性化材用はいずれもneomycin Bの併用によって抑制された。(5)細胞内外のカルシウムキレ-ト剤BAPTA‐AMおよびEGTAの併用はBK、B_1、およびArgのカテプシンB活性化作用をいずれも抑制した。(6)各種プロテインキナ-ゼの阻害剤、H‐7、W‐7、およびH‐8の併用はBK、B_1、およびArgのカテプシンB活性化作用を抑制した。(7)線維芽細胞にBK、B_1、およびArgを1分間作用させた後、阻害剤に置き換え、カテプシンB活性維持の時間経過における阻害剤の影響を検討した。neomycin BはいずれもカテプシンB活性を適用直後より消失させた。H‐7、W‐7、およびH‐8はB_1刺激後のカテプシンB活性維持に対して1分、4分、および4分後より抑制作用を示したが、BK刺激後のカテプシンB活性はいずれも4分後において抑制作用が認められた。Argではいずれの阻害剤も刺激後1分において抑制作用が認められた。
