急速凍結・電顕法を用いた生体分子のミリ秒時間分解能における動態分析

研究概要	caged化合物とは、強い紫外線があたると分解してcageがとれ、活性型になるように合成された化合物のことである。その代表的なものにcaged ATPがある。この化合物を含む溶液に強い紫外線をあてると、溶液中のATP濃度を急激に上昇させることが出来る。したがって、このような化合物を含む生物試料を凍結の直前に(ミリ秒のレベルで正確に)強い紫外線にあてる装置ができればその利用価値は大きい。しかし、このような装置を作る場合、解決されなければならないいくつかの技術的な問題点がある。本年度は、これらの問題点を一つずつ解決し、caged化合物を用いた急速凍結システムを開発することを目指した。まず、第一にいかに強い光源を得るかということ。レ-ザ-光線を用いることも考えたが費用の点から難しく、従来のランプ形の光源を改良して充分なエネルギ-が得られるように工夫した。第2に光源の光をどのようにして試料まで導くかということ。鏡を使うことも考えたが光軸を合わせることが難しい。紫外線用のライトガイドを凍結棒に組み込むことにより解決した。第3に生物に有害な波長の光を除くこと。フィルタ-をうまく組み合わせることにより除去できた。第4に光をあてるタイミングと凍結のタイミングを正確にかつ自由に設定できること。光のセンサ-とコンピュ-タ-を組み込むことにより解決した。このような問題点を解決することにより、充分なエネルギ-の無害な光を凍結の直前5ミリ秒から200ミリ秒の間に正確に試料に照射できるシステムが完成した。さらにこのシステムには、試料が凍ったときに電導度が変わることを利用して凍結の良い悪いをその場で判定できる装置も組み込まれた。現在、このシステムを用いて筋収縮の基礎メカニズムについて解析を進めている。
