巨大DNAクロ-ニングによるヒトHLA抗原遺伝子の構造解析

研究概要	ヒトHLA遺伝子領域に位置する疾患感受性遺伝子の同定、遺伝的多型性の生成の機序の解析、ヒトの遺伝子構成の解明などを目的として、我々はHLA全領域のクロ-ニングを目指している。我々のグル-プはすでに全3.500kb中1.450kbをコスミドベクタ-にクロ-ニングしていたが、今年度はコスミドに加えてYACベクタ-によるクロ-ニングの系を確立し新たにクラスI抗原領域を中心に800kbをクロ-ニングした。YACクロ-ンのインサ-トサイズは160kb〜700kbであり、HLAーB及びC遺伝子周辺に数多くのCpGislandが存在することが明らかとなり、これらの遺伝子とクラスI抗原と相関する多くの疾患(ベ-チェット病、尋常性乾癖、強直性脊椎炎など)の原因遺伝子との関係を追及するために、現在CDNAクロ-ンの分離、解析を行っている。またクラスIII遺伝子よりクラスII遺伝子に向けてコスミドウォ-キングをおこない、150kbの領域をクロ-ニングした。この領域はGCrich→ATrich領域の変化する興味ある部位であるが、コスミドクロ-ンの解析によりこの変化が連続的ではなく、不連続的であることが示唆された。またこの解析の過程で21ーOHハイドロキシラ-ゼ遺伝子より約100kbセントロメア側にテネイシン様の遺伝子が塩基配列の決定によりみいだされた。すなわち、N末端から中心球状部、7反復部、EGF様繰り返し配列、フィブロネクチンIII型繰り返し配列、フィブリノ-ゲン様配列が続く典型的なテネイシン様構造をもつことが明らかとなった。なお興味あることに、このテネイシン様遺伝子と21ーOHハイドロキシラ-ゼB遺伝子の3'末端は互いにオ-バ-ラップしていた。この遺伝子とクラスII抗原と相関する疾患との関係は今後の課題である。
