合成キノンの光合成反応中心Iへの再構成ーキノン結合部位の立体構造とエナジエティクス

研究概要	ホウレンソウ葉緑体の光化学系1標品をエ-テル抽出し、初期電子受容体として働くフィロキノン(VK1)を除いた反応中心を得た。これに各種キノン、阻害剤を再構成し、改造反応中心を作成し、内部電子移動過程をマイクロ秒分光法、ナノ秒時間分解スペクトル法、極低温ESR法等により解析し、以下のように初年度の研究計画をほぼ達成した。 1)再構成キノンの立体構造と、反応中心1への結合力の関係をあきらかにし、キノン結合部位の立体分子構造を推定した。キノンの反応部位への結合は、キノン分子の疎水性・立体構造・カルボニル部の水素結合形成能等で規定されることをあきらかにした。各種キノン誘導体の結合定数を決定し、その構造との比較からキノン結合部位の構造を推定した。 2)合成キノン再構成後の反応の解析によりキノン結合部位の特性を解明した。置き換えキノンが還元力をクロロフィルから鉄硫黄センタ-(FeS)に伝える効率から、その実効酸化還元電位を推定し、有機溶媒中での値と比較した。この結果光化学系1反応中心のキノン結合部位は非常に強い還元力を作り出せる反応環境を作っていること、及び、この特性は、これまでに立体構造があきらかにされた紅色光合成細菌反応中心や光化学系2のキノン結合部位とは大きくことなることが示された。 3)阻害剤のキノン結合部位への結合ー新たな除草剤結合部位の発見。キノン結合部位に働く阻害剤のうち、アトラジン型の阻害剤(紅色細菌、光化学系2反応中心には有効)は光化学系1には効かず、フェノ-ル型の除草剤は効くことが明らかになった。光化学系1反応中心に結合する阻害剤の報告はこれまで全く存在せず、我々の報告が最初のものである。
