合成キノンの光合成反応中心1への再構成

研究概要

高等植物葉緑体の光化学系1反応中心複合体に存在するフィロキノンを、他の機能を損なう事なく抽出し、再構成出来ることを多くの合成キノン及びその類似体で示した。これにより、動物の血液凝固に必要なビタミンK1(フィロキノン)の植物中での本来の役割が、この反応中心での電子受容体である事を明らかにした。これは、これまで鉄硫黄センタ-が電子受容体として働き、キノンは働いていないとされていた光化学系1反応中心でも、光化学系2あるいは紅色細菌型の反応中心と同様にキノンが2次電子受容体として働くことを示すもので、光合成反応中心の進化、起源を考える上でも重要な発見である。
このフィロキノン結合部位には本来のフィロキノンのみでなく、異なった性質、構造を持つ百種以上の合成キノンあるいはキノン類似化成物、除草剤を導入出来る事を明らかにした。この部位に導入された化合物は、フィロキノンと同様の電位を有する化合物のみが本来の電子移動反応経路を完全に回復することを明らかにした。キノンの構造、性質と反応特性を詳しく検討し、反応中心蛋白質はいくつかの基本的性質を満たす外部導入化合物に対しては非常に効率のよい反応環境を与えることを明らかにした。光化学系1反応中心蛋白質中にいれられたキノンは有機溶媒中よりもずっと強い還元力を示し、一方、同じキノンを光化学系2反応中心あるいは紅色細菌反応中心にいれると、有機溶媒中よりも弱い還元力を示すことから、キノンの構造のみでなく、蛋白質とキノンの相互作用が非常に重要な事も明らかにした。これらの事実から、光合成反応中心蛋白質が電子移動の反応環境として高い自由度をもち、進化の過程で構造を変えて、生物の必要に応じた性質を作り出してきた事が明きらかになった。
