ウニ受精卵有糸分裂の分子機構(染色体はどう動くか?)

研究概要

ウニ受精卵を10^<-1>Mβ-メルカプトエタノ-ルで第1分裂前期から11/2細胞周期処理すると、正常海水に戻された処理卵は直ちに紡錘体2個を形成し、4割球となる。次に各割球に形成されるのが半紡錘体であって、正常紡錘体の半分の大きさを持ち、且つ複屈折量は等しいから、半紡錘体を構築する微小管数は等しい。然し、反対極側の動原体微小管を欠くこの半紡錘体では、後期染色体運動は殆ど認められなかった。これは一つには適当な解析用記録の方法が無かった事による。そこで私達は当菅島臨海実験所で試験開発した超顕微鏡と、VTR画像記録装置を用い、半紡錘体の形成と脱構築過程を偏光およびノマルスキ-微分干渉法によって連続記録し、染色体運動の有無について検討した。その結果、1)半紡錘体は長く中期に留めるが、殆ど後期染色体運動を示さず終期に入り、染色体は中心体周辺に集まった核様体となる。2)然し、長く中期に留った後に娘染色体は核板上で分離するから、後期に入るのは確実である。但し染色体は分かれるが極へは移動しない。3)第一分裂で卵割しそこなった卵では、次に4個の半紡錘体が形成される。この時2個の半紡錘体が融合して正常なみかけの紡錘体が作られるが、この紡錘体はコントロ-ルと同様なタイミングで分裂する。残りの2個の半紡錘体も、みかけ正常な紡錘体と同調した複屈折性の消長を示す。しかし、後期染色体運動は起こらない。4)中期半紡錘体に40%D_2Oを添加すると、反対極側娘染色体の複屈折動原体系が形成され、明らかに娘染色体が移動する。但し動きの方向はばらばらである。これはかなめとなる中心体が欠失している為と考えられる。以上の観察は後期染色体運動に極中心体は必要ではないとする平本やNicklasの動原体モ-タ-説やパックマンモデルに反するものであり、分裂後期における極中心体間の動的平衡の重要さを支持するものである。
