魚類における産卵雌特異物質とビテロゲニンの挙動

研究概要	本課題のもとで行った研究は下記の1および2より成り、それぞれの結果を記す 1産卵雌特異物質(SF物質)とビテロゲニン(Vg)の肝臓内合成:メダカより単離精製した両物質のそれぞれと特異的に反応する抗体を作製し、雄成魚肝臓切片および肝臓抽出液を用いて免疫組織化学法および免疫ブロット法によって、エストロゲン投与後の両物質の生成過程をしらべた。両物質はともに投与後8〜12時間後に検出され、以後時間を追ってほぼ平行して増加してゆくことがわかった。これにより、両物質の遺伝子はエストロゲンの作用で可成り短時間のうちに、しかも殆んど同時的に発現することが予想される。本研究の過程で、SF物質に特異的に反応する抗一卵膜糖タンパク質抗体と交叉反応をする複数の高分子タンパク質が新たに発見され、従来のSF物質との識別や精製を行うことになったので、肝臓からのSF物質やVgのmRNAの分離は未だ行っていない。 2卵内のVgおよび卵黄タンパク質の発生に伴う変化:発生初期のメダカ胞(胞胚)卵黄リンタンパク質をポリアクリルアミドゲル電気泳動法によりしらべ、2種の高分子量タンパク質と2種の低分子量タンパク質を見出している。前2者(PP1およびPP2)はVgと似たアミノ酸組成を示し、免疫ブロット法により抗一メダカVg抗体と反応することがわかっている。後2者(PP3、84KDaおよびPP4、47KDa)はリン含量やアミノ酸組成から、メダカのフォスビチンと考えられる。PPー3とPP4はそれぞれ単離精製した。以上4種類のリンタンパク質は発生第1日目と第2日目の間に急速に脱リン酸され、タンパク質部分も分解される。この変化は30℃で孵卵すると受精後10時間目の胚で認められはじめ、酸性フォスファタ-ゼのほかにプロテア-ゼの関与が示唆されている。
