植物病原糸状菌への遺伝子導入と形質転換ベクタ-系の開発

研究概要	本年度に実施した研究の概要を以下の項目別に要約する。 1.植物病原菌胞子に外来遺伝子を直接導入するための実験法の確立植物病原菌としては、トマト萎凋病菌の小型分生胞子を使用し、エレクトロポ-レ-ション(EP)法によって外来高分子の導入を試みた。EPの条件としては、電場強度、パルス回数、導入物質の濃度を検討し、最適条件を明らかにした。これらの研究結果は、Journal of Phytopathology誌の1989年第125巻89〜96に掲載された。また、本法を用いた外来遺伝子の導入については、水銀無毒化遺伝子を組込んだベクタ-DNAを使用し、病原菌胞子染色体に挿入され、かつその遺伝子の発現が認められることを明らかにした。これらの研究結果については、平成元年度日本植物病理学会大会(札幌)で発表した。 2.糸状菌で作用するプロモ-タ-領域の探索トマト萎凋病菌をデンプン添加培地で培養したところ、活発なアミラ-ゼ王産が認められたので、アミラ-ゼ王産遺伝子の転写、すなわち本遺伝子のプロモ-タ-活性が強力であると考え、アミラ-ゼ遺伝子の単離とその上流部塩基配列の解析を試みることにした。まず、萎凋病の染色体DNAから遺伝子バンクを作製する目的で、Sai3AI制限酵素で切断したDNAをEmbel3ファ-ジベクタ-に組換え、染色体全部をカバ-するクロ-ンを分離した。次に、アミラ-ゼ遺伝子を単離するため、デンプン添加培地で培養中の菌体からmRNAを分離し、上述の遺伝子バンクと相同性試験を行った。今回の実験によってアミラ-ゼ遺伝子を分離したので、これらの結果を学会発表するとともに、さらにそれらの遺伝子構造を解析する予定である。
