微生物における環境変化に応答した情報伝達と遺伝子発現制御

研究概要

大腸菌は外界の浸透圧変化に対して様々な応答をする。本研究で対象とした大腸菌外膜タンパク質OmpF,OmpCの合成制御を見てみると、高浸透圧条件下では一方のタンパク質(OmpC)が優先的に合成され、低浸透圧条件下では他方(OmpF)が優先的に合成される。外界の物理的要因への細胞応答やそれに伴った遺伝子発現制御という観点から、この一見奇妙な制御メカニズムに関心を持ち、分子生物学的な研究を展開してきた。本研究において我々が明らかにしてきた要点を以下にまとめる。(1)、OmpF、OmpCの合成制御は遺伝子の転写のレベルでなされいる。(2)、この転写制御には少なくとも二つの調節タンパク質(OmpR、Envz)が重要な働きをしている。(3)、OmpRはompF、ompC両遺伝子のプロモ-タ-上流DNAに結合し、転写を促進する遺伝子活性化タンパク質である。(4)、EnvZは細胞質膜に存在し、浸透圧を関知するセンサ-タンパク質と予想され、浸透圧変化の情報をなんらかの機構でOmpRに伝達する機能を担っていると考えられる。
本年度の研究計画における特に大きな成果は、EnvZの機能を具体的に明らかにすることができたことである。その成果を以下にまとめる。(1)、EnvZは膜局在性のタンパク質リン酸化酵素であり、自己リン酸化能を有する。(2)、EnvZはOmpRを特異的にリン酸化する。(3)、EnvZはリン酸化型OmpRの脱リン酸化にも必須である。(4)、OmpRはリン酸化されることによりDNAへの結合能が上昇し、ompF、ompC遺伝子の転写を顕著に促進する。(5)、以上の結果を基にEnvZによるリン酸化を介した情報伝達と遺伝子発現制御に関する分子レベルでのモデルを提唱した。本研究におけるこれらの発見は、微生物における情報伝達機構及び遺伝子発現制御機構にタンパク質のリン酸化が関与している事を実験的に示した例として、この分野の発展に大きく貢献することができた。
