内分泌細胞によって形成されるコロイドを含む小濾胞の研究

研究概要

モルモット下垂体に存在するコロイドを含む小濾胞について、PAS染色、各種下垂体ホルモンと濾胞細胞に特異的に反応するSー100蛋白の抗体を用いての免疫組織化学および電顕で調べた。隆起部では3種類のコロイドを含む小濾胞が観察される。すなわち濾胞細胞によって形成される典型的な濾胞、空胞状物質(膨大した粗面小胞体の槽)で充満した細胞によって形成される濾胞、および性腺刺激ホルモン(LHとFSH)分泌細胞によって形成される濾胞である。隆起部では濾胞は生後5日令の幼若動物にも存在するが、年令と共に増加し、24ケ月令以上のモルモットでは5日令の10倍以上増加した。前葉においてもコロイドを含む小濾胞が観察される。しかし隆起部とは異なり幼若動物には全く存在せず、6ヶ月令から濾胞が形成され始め、以後年令と共に増加し、24ヵ月以上のモルモットでは著しく多くの濾胞が存在する。水平断した前葉の4切片の合計は6ヶ月令で雄1.2±0.7、雌3.5±0.5であるが、24〜36ヶ月令では雄410.5±92.3、雌454.7±84.7であった。また濾胞腔に貯えられるコロイドの直径は老令動物では6ヶ月令の2倍以上増加していた。このようにモルモット下垂体前葉でコロイドを含む小濾胞の形成は老化現象によることが解明された。前葉の濾胞を形成する細胞は主にSー100蛋白の免疫活性を示し、また電顕による微細構造上からも濾胞細胞であることが確認された。濾胞細胞の他に成長ホルモン(GH)分泌細胞が濾胞腔のコロイドと接していた。老令動物ではGH細胞の合胞体が形成されるが、GH細胞合胞体の細胞質内にコロイドや線維状物質を貯える小腔が存在した。ATCH細胞の集団、TSH細胞、LHおよびFSH細胞も濾胞腔に接する像が認められた。モルモット下垂体前葉では濾胞は主に濾胞細胞によって形成されるが、ホルモンを分泌する細胞をミコロイドと直接接し、濾胞の形成に関与すると考えられる。
