前庭頸射における単一前庭脊髄路細胞による頸筋支配の空間パタ-ンの解析

研究概要	頭部の偏位を補償する前庭頸反射は、頭部の偏位の起こった平面で逆方向に起こることが知られ、これは三半規管が受容器として働いている。この前庭頸反射の神経回路は、半規管ー前庭神経ー前庭神経核ー前庭脊髄路ー頸筋運動ニュ-ロンよりなることが明らかにされている。頭部の運動を引き起こす頸筋は20以上あり、三半規管からの出力がどのような機構でこれらの頸筋支配の空間パタ-ンを形成しているかは運動制御の重要な問題である。本研究は、単一半規管入力を受ける前庭脊髄路細胞が脊髄において複数の筋を支配し、この空間パタ-ン形成に関与している可能性を調べることを目的をしている。ネコにおいて、上部頸髄から軸索スパイクを記録し、回転台を用いていづれの半規管入力を受ける前庭脊髄路細胞であるかどうかを同定した後、ホ-スラディシュパ-オキシデ-スを電気泳動的に注入し、連続染色切片を用いて、軸索分枝の形態を解析した。単一前庭脊髄路細胞は、複数の軸索側枝を持ち、脊髄のVII,VIII,IX層に主に分布していたが、IX層では、腹内側核と副神経脊髄路核に投射していた。1/3の細胞がこの両運動神経核に投射しておりホ-スラディシュパ-オキシデ-スでラベルされた運動ニュ-ロンの細胞体又は近位樹状突起にシナップス結合をしていた。腹内側核は、固有背筋の運動核であるが、この中に複数の運動核があり、単一前庭脊髄路細胞の分枝は、複数の核に同時に投射していることが明らかとなった。以上の結果は、単一前庭脊髄路細胞が複数の筋の興奮性を同時に支配したおり、補償性の前庭頸反射を引き起こすに必要な頸筋群の空間的収縮パタ-ンの形成に関与していることを強く示唆するものである。この分枝パタ-ンは、興奮性及び抑制性前庭脊髄路細胞のいずれにも見いだされた。
