小腸レチノ-ル結合たん白質Type IIの生理作用に関する研究

研究概要

1.ラット小腸のLーRAT活性とCRBP(II)量に及ぼす栄養条件の影響:18日齢の幼若ラット、または5週齢のラットをビタミンA無添加食で14日間飼育した場合、また5%カゼイン食と20%小麦グルテン食で14日間飼育した場合、LーRAT活性は有意に低下した。しかしながらCRBP(II)量は変動しなかった。従って、レチノ-ルのエステル化酵素(LーRAT)は、食餌中のビタミンAの有無と蛋白質の質と量の影響を受けるが、そのエステル化で担体の役割をもつGRBP(II)量はそれらの影響を受けず腸管から入ってくるレチノ-ルを受け取るに十分な量が常に用意されていることが明かとなった。
2.ラット小腸CRBP(II)のcDNAプロ-ブの作製:ラット小腸粘膜からmRNAを抽出して、cDNAを合成した。この合成に当り、報告されているCRBP(II)mRNAのヌクレオチド配列をもとにしてCRBP(II)mRNAの5'末端と3'末端のオリゴヌクレオチドを合成し、polymerase chain reaction(PCR)によってcDNAプロ-ブを作製した。
3.栄養条件によるCRBP(II)発現変動の解析:離乳直後のラットをビタミンA欠乏食により長期(2ー3カ月)間飼育して作製したビタミンA欠乏ラットの、さらにその欠乏ラットにビタミンAを再投与されたラットの空腸から総mRNAを抽出し、完成させたCRBP(II)のcDNAを用いて、mRNAの定量を進行中である。cDNAプロ-ブの完成が予定より遅れたため研究成果も遅れている。
4.小腸組織におけるLecithin:retinol acyl transferase(LーRAT)の局在の解明:ラットLーRNTの抗体作製を兎を用いて進行中である。その抗体の完成を持って、小腸組織でのLーRATの局在の免疫組織学的に解明する。
