プロスタグランジンF合成酵素の構造と機能

研究概要	1.プロスタグランジン(PG)F合成酵素 PGF合成酵素は、分子量36,666の単一鎖の蛋白質で、NADPH存在下に異なる活性部位でPGH_2およびPGD_2を還元し、それぞれのPGF2αおよびPGF2αの立体異性体で生理活性の異なる9α,11βーPGF2を生成する酵素である。昨年度までの研究に於て、本酵素は少なくともに二つのドメインからなり ^<195>ProがPGD_2の活性部位に関連することを明らかにしてきた。本年度は、二つの活性部位を更に明らかにするためにPGD_2の誘導体を用い本酵素を光親和ラベルし、PGD_2の活性部位に関与するいくつかのペプチドを決定することが出来た。また、cDNAの3'末端からの削減で、本酵素のC末端から100残基のアミノ酸は、PGD_2の還元に関与しないことが明らかになった。 2.ウシ肝臓PGD11ケト還元酵素の精製とその性質上記のPGF合成酵素に対する抗体を用いてPGF合成酵素の組織分布を調べた結果、肝臓に本抗体と約80%交差し、PGD_2のKmがPGF合成酵素やウサギ肝臓PGD11ケト還元酵素と異なる新しいタイプのPGD11ケト還元酵素が存在することが明らかになった。そこでウシ肝臓よりPGD11ケト還元酵素を均一にまで精製し、その性質を明らかにした。その結果は、精製酵素においてもPGD_2のKm値は10μMと低いものの、PGD11ケト還元酵素もPGF合成酵素と同様に広い基質特異性を示し、種々のケト化合物やPGD_2のみならずPGH_2をも基質にすることが明らかになった。更にPGD_2を含むケト類の還元活性は、クロライドイオンで活性化されるが、PGH_2の還元活性は影響が無いことから二つの活性部位の存在が示唆された。以上の結果、PGD11ケト還元酵素もPGF合成酵素と同様に多機能酵素であることが明らかとなり、これらの酵素が多機能酵素の構造を機能を明らかにする一端となると考えられる。
