毒素原性大腸菌耐熱性下痢毒素の受容体とグアニレ-トシクラ-ゼ活性化機構

研究概要

STh受容体の精製と構造解析を中心に研究を進めた。
ラットの小腸上皮細胞から刷子縁細胞膜を調製し、各種の界面活性剤を用いてSTh受容体の可溶化を試みた結果、0.1%Lubrol PXがもっともSTh受容体を効率よく可溶化することがわかった。そこで、Lubrol PX処理して得られた粗STh受容体を濃縮後、Sephacryl S-300HR カラムクロマトを行った。分子量200,000を示す画分にフォトアフィニテイラベリングでSThと結合する。3種類の蛋白が検出れた。この3種の蛋白のSDS-polyacrylamide電気泳動により求まる分子量は53,000、70,000、77,000であり、ゲル濾過法によって求めた分子量の値200,000より小さく、STh受容体がいくつかのサブユニットから抗生されていることを示唆していた。ここで得たSTh受容体画分をCon A Sepharoseカラムクロマト行いさらに精製したところ、0.5M α-methylglucosideと1.3M α-methyl-mannosideによって溶出されるSTh受容体とに分離された。そこで最もSThと強く結合する70kDa蛋白を含む画分、1.3M α-methyl-mannoside画分に溶出されたSTh受容体をSTR-200Aとし、0.5M α-methylglucosideで溶出されたSTh受容体をSTR-200Bとした。STR-200BはSThと結合する二つのサブユニット(53kDa蛋白と77kDa蛋白)を有していた。従ってラットの腸管膜のSTh受容体は2種類ありいずれも糖蛋白であることが示唆された。興味深いことにSTR-200AとSTR-200Bの糖鎖は異なっており、STR-200Aの糖鎖はN-linked high mannoseタイプで、STR-200Bの糖鎖はN-linked complexタイプであった。現在STR-200Aの3本の糖鎖はSThとの結合に関与しないことがわかっている。またSTR-200AとSTR-200Bにはグアニレイトシクラ-ゼ活性は検出されていない。STh受容体精製の方法としてSThの活性アナロ-グにビオチンを3分子結合させたリガンドを作成し、これを用いたアフィニテイクロマトが有効であることがわかった。
