主要組織適合性抗原クラスエ遺伝子の転写制御と発癌・臓器移植

研究概要

アデノウィルス12型EIAによる組織適合性抗原クラスエ遺伝子の転車抑制に関与するシス、エレメントを、DNaseIフットプリント、ゲルシフト法で決定し、プロモ-タ-の2kb上流のTandemに並んだCAAリピ-トに同定した。このエレメントをクラスエcDNAのプロモ-タ-上流に挿入すると、顕著に、Hー2クラスエ抗原の発現がEIA依存的に抑制することを、FACS解析で明らかにした。更に、このCAAリピ-トに結合する核内蛋白質、分子量74 Daと58kDaをUVクロスリンク法で同定した。しかしながら、このエレメントは、MHC内の全てのクラスエ抗原遺伝子のプロモ-タ-上流に存在するのではなく、b及びdハプロタイプ共に、Hー2KとQa遺伝子の一部にしか見い出されない。そこでこのCAAリピ-トをプロモ-タ-にこのエレメントが存在していないHー2遺伝子の上流に挿入すると、見事に、その発現が抑制される事より、このCAAリピ-トは、MHC複合体のクラスエ遺伝子のEIAによる発現抑制に必須のものである可能性が極めて高い。その他、クラスエ抗原遺伝子のBasalな転写制御に関与する正と負のエレメントを従来のエンハンサ-CRE/ICSの領域以外に見い出し、その活性の検索を行なっている。正の領域は、プロモ-タ-上流ー370よりー292の内に存在し、Heat Shock ConcensusとPurine boxの配列が連続して存在している。これらの配列には、3種類の異なった蛋白質が結合する。負の領域は2箇所存在し、(1)ー294よりー213(2)ー560よりー502の配列である。これらの領域には共通の蛋白質が結合している可能性が高い事を、ゲルシフトの競争実験で明らかにした。その他、正のエレメントには、癌抑制遺伝子産物Rbが結合する配列が存在し、今後、クラスエ抗原と発癌との相互関係に新しいロ-ジックを導入できる期待がかかっている。
