研究概要

1.前年度に引き続き、遺伝的多型を示すリボヌクレア-ゼ(RNase)の起源や分布を知るために各種臓器からRNaseを精製し、その性状を比較検討した。硫安分画および一連のカラムクロマトグラフィ-を利用し、ヒト腎臓から分子量の異なる3種類のRNaseを精製し、RNase HKー1,HKー2A,HKー2Bと命名した。これらについて、アミノ酸組成やN末端アミノ酸配列などの蛋白質化学的、酵素学的および免疫学的諸性状を調べたところ、RNase HK1は尿Nonsecretory RNaseに、一方RNase HKー2AとHKー2Bは尿secretory RNaseに類似していた。しかし、RNase HKー2AおよびHKー2Bと尿secretory RNase、RNase HKー1と尿nonsecretory RNaseのそれぞれの糖組成間には明らかな相違が見られた。従って、尿RNaseの起源は腎臓ではないものと考えられた。
2.前年度、我々は尿デオキシリボヌクレア-ゼI(DNase I)に遺伝的多型のあることを明らかにした。本年度は、遺伝的多型を示す尿DNase Iの生化学的性状を精査し、さらに血清DNase Iについて人類遺伝学的解析を行った。(1)一連のカラムクロマトグラフィ-を利用して、単一個人より採取した尿からDNase Iを精製し、アミノ酸や糖組成、N末端アミノ酸配列などの蛋白質化学的および酵素学的樋状を明らかにした。特に、尿DNase Iの分子多様性は一次構造上の置換およびシアル酸含量の差に起因するものと考えられた。(2)血清DNase Iについて等電点電気泳動法と新たに開発した高感度活性染色法を併用することによって、血清DNase Iアイソザイムを再現性よく分離・検出することに成功した。日本人約500人の血清DNase Iを解析したところ、血清DNase Iは少なくとも10種類の表現型に分類することができた。家族調査などの詳細な人類遺伝学的解析から、血清DNase Iは共優性な少なくとも4個の対立遺伝子によって決定されていることを明らかにした。
