研究概要

1)我々はこれまでに血清リボヌクレア-ゼと尿デオキシリボヌクレア-ゼI(DNase I)とは、遺伝的多型形質であることを明らかにしてきた。本年度は、ヒト体液中に広く分布しているもうひとつの核酸分解酵素DNase IIについて検討した。ヒト尿から高純度なDNase IIを分離し、その生化学的性状を精査した。得られた知見を基礎に、高感度な活性測定法や等電点電気泳動法(IEFーPAGE)と、新たに開発した活性染色法との併用によるアイソザイム検出法を確立した。同一個人より採取した尿、白血球および精液中のDNase II活性強度には、互いに密接な相関がありしかも明白な個人差が見られた。日本人約600人の集団調査から、尿DNase II活性強度は二峯性分布を示し、2種類の表現型(高活性型と低活性型)に分類できることがわかった。家族調査より、尿DNase IIは常染色体上の一対の対立遺伝子によって決定されており、低活性型は劣性形質であることも明らかにした。さらに、白血球DNase IIに関しても尿DNase IIと同様な遺伝学的結果が得られた。
2)ヒト血清中に存在する凝固・線溶系の一因子であるプロテインCインヒビタ-(PCI)について、抗PCI特異抗体を用いた免疫ブロット法とIEFーPAGEを併用することによって、血清中PCIイソ型タンパク質を再現性よく分離・検出することに成功した。日本人約1000人について血清中PCIを解析したところ、血清PCIを3種類の表現型に分類することができた。家族調査から、血清PCIは常染色体性相互優性な2個の対立遺伝子によって支配されていることを明らかにした。さらに、血清PCIのN末端およびC末端側のアミノ酸15残基に相当するペプチドに対するそれぞれの特異抗体を用いた免疫ブロット法でも、抗PCI特異抗体によるものと同一のPCIイソ型タンパク質パタ-ンが観察され、血清PCI解析における抗ペプチド抗体の有効性が知られた。
